Course Objective

To get know about basic recombinant DNA technologies and bioinfirmatics; to be able to reach and analyse in silico DNA, RNA and protein data; to understand and apply basic DNA, RNA and protein techniques

Learning Outcomes of the Course Unit

1   To be able to discuss about human genome project and its outcomes; to explain HapMap and 1000Genomes projects
2   To be able to use bibliographic databases such as Pubmed ve OMIM; to be able to reach gDNA, cDNA, exon-intron sequences, protein sequence of a certain gene by using Entrez and Ensemble Genome Browsers.
3   To be able to align nucleic acid and protein sequences by using CLUSTAL W program; to know GEO, UniProt and KEGG tools
4   To be able to understand basis of gene cloning and recombinant DNA technologies
5   To be able to isolate gDNA, total RNA and total protein lysate from the cell and to make it quantify.
6   To be able to discuss basic properties of plasmid vectors and types of plasmids used in different cloning purposes; to be able to isolate plasmid from bacteria; to understand different forms of plasmid by using restriction enzymes
7   To be able to analyse gen expression at transcript level by using semi-quantitative RT-PCR and at protein level by using western blotting and immunostaining
8   To be able to optimize and apply conventional PCR reaction and design primers
9   To be able to analyse and present data

5 DNA replication, gene expression (transcription and translation) and their role in disease
6 Types of chromosomal abnormalities
7 Mutation and polymorphism, Types of single gene mutation and single gene disorders
8 Mechanisms of DNA damage, DNA repair and related disease models
9 Midterm exam
10 Identification of disease and disease-related genes
11 Genetic and epigenetic control mechanisms
12 Cell- cell and cell-ECM interactions, microenvironment and their role on the disease progression
13 Molecular basis of cancer
14 Definition of Stem cell, types of stem cells and their usage in medicine
