It was aimed to give students basic information and principle of cell culture which is one of the current research fields.

Learning Outcomes of the Course Unit

1   Have knowledge about the importance, basic terminology and principle of cell and tissue cultures.
2   Have knowledge and generate ideas as to the space requirements, equipments and materials used in cell culture laboratories.
3   Have knowledge about basic techniques, media, sterilization techniques and the factors caused to contamination and apply them.
4   Have knowledge about microscopic imaging techniques and analyze the images.
5   Have knowledge about subculturing of primary and special cell cultures, cell cloning and scale enlargement in cell culture.
6   Have knowledge about the technological applications of cell culture.

Course Contents

1 Historical background and introduction to basic terminology of cell and tissue cultures
2 Setting up a cell culture laboratory; space requirements, equipments, culture plasticware and associated small consumable items, maintaining the laboratory
3 The physical environment; temperature, pH, osmolality, CO2 and other gases Surfaces and cell shape; adherent versus nonadherent cells
4 Sterilization; wet heat, dry heat, irradiation, chemical sterilization, filtration
5 Cell culture media; basic components of the cell culture environment, preparation of culture media
6 Standard cell culture techniques; subculturing of adherent and suspension cultures, growth curves and measuring cell growth, freezing and thawing cells
7 Microscopy of living cells; general imaging methodology, imaging of intact cultured cells in flasks, dishes and plates, high-resolution imaging of cells on coverslips, imaging of fluorescently labelled cells, recording of microscopy images, analysis of microscopy images
8 Midterm
9 Contamination; avoid and recognize, method for fluorescent detection of mycoplasma, cross-culture contamination
10 Development and optimization of serum and protein free culture media
11 Primary culture and the establishment of cell lines
12 Specific cell types and their requirements
13 Cell cloning procedures; choice of technique, methods for attached and suspension cells
14 Cell culture for commercial settings; the cell as industrial property, small and large scales

Meaningful learning of the basic concepts given in presentations, association of concepts with each other, establishing the cause-result relationships and making comments by using the information available for problems and evaluating idea generation are carried out with mid-term and final exams, active participation to the lecture, homework.
