Course Objective

Objectives of this course are:
*To acquire basic knowledge on cell culture techniques
*To gain an understanding of the biology and adaptation process of culture cells
*To perform basic cell culture techniques

Learning Outcomes of the Course Unit

1   Outlines the biological features of cells in tissues and cells in culture
2   Recounts the basic knowledge on cell proliferation, transformation, cell death and differentiation
3   Knows the equipment, consumables and culture media used in the cell culture laboratory
4   Knows and practises bio-safety rules, aseptic technique and sterilization procedures in the cell culture laboratory.
5   Describes cloning and transfection techniques
6   Describes cell characterization and separation techniques
7   Prepares culture media; seeds cells in culture; maintains cells in culture; passes/splits suspension and monolayer cultures; freezes and thaws cells; and performs basic viability tests.
8   Monitors culture cells daily and keeps his/her laboratory notebook accordingly.

3 Preperation of cell culture media and cell seeding Sterilization procedures, aseptic technique and bio-safety in the cell culture lab
4 Features of primary culture cells and cell lines Splitting suspension culture cells, counting cells and viability tests
5 Contamination in cell culture Freezing protocols Monolayer culture seeding
6 The concepts of transformation and transfection Preparation of freezing media and freezing suspension culture cells
7 Cell cloning, separation and selection methods Splitting monolayer culture cells
8 Characterization of culture cells Differantiaiton of cells in culture Thawing cells
9 Project work on HeLa cell line (discussions and application)

Planned Learning Activities and Teaching Methods

Lectures: Basic theoretical concepts will be explained during lectures
Homework: Students are expected to hand in 4 written reports on topics that are given by the course coordinator.
Project: Each student will monitor, split, maintain, freeze, thaw and perform viability tests on a Hela cells given by the course coordinator during the course. Every observation and application is expected to be recorded in his/her laboratory notebook; which will be handed in at the end of the course.

Further Notes About Assessment Methods

Learning objectives 1-6 will be assessed by a written mid-term exam and by the evaluation of written reports handed in as homework.
Learning objectives 7 and 8 will be assessed by the evaluation of the laboratory notebook handed in by the student; as well as direct observation of his/her lab work. Attendance will also be taken into consideration.
Grading (%):
Mid-term exam: 40%
Homework Assignments/Presentation: 10%
Project & Laboratory work: 50%
