Course Objective

The purpose of this course is to teach basic laboratory principles and general laboratory procedures and also to introduce separation techniques. The course consists of theory and practice sections. This course is given as a mini project. In theoretical section of the course, the basic principles and procedures regarding the method are given.
In the implementation stage of the course, the supernatants are obtained from rat tissues (brain, liver, heart, kidney, lung) by weighing, homogenization and centrifugation. The supernatants obtained are used in all methods to learn throughout the semester. In these supernatants primarily the protein concentrations are determined using spesific protein methods like Biuret, Modified Lowry and Bradford.
Then, Glutathione-S-transferase and MDA levels are measured by using a spectrophotometer, spectrofluorimeter and Liquid Chromatography (HPLC) method, respectively. The molecular weights "MW" of unknown protein bands are calculated by SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis).The is Students make presentations related with their results in the end of the semester.

Learning Outcomes of the Course Unit

1   Be acquired the knowledge about the basic biochemistry and basic chemistry which are the main areas of biochemistry.
2   Be grasped the theoretical knowledge about the basic biochemistry and basic chemistry which are the main areas of biochemistry.
3   Have the knowledge about the principles and methods of basic laboratory equipments.
4   Be able to critically analyze the basic concepts of basic laboratory methods (eg, such as differences in methods of protein determination)
5   Synthesis: Be able to discuss the learned methods by considering different ways and be able to adapt to new situations (Example: to be able to discuss why different tissues contain different proteins)
6   Be able to present by explicating the results of one of the applied methods.
7   Be acquired the basic laboratory skills ( for example: to weigh, buffer preparation, pH measurement, to make homogenize, to make centrifuge)
8   Be acquired the application skills of the general laboratory prosedures
